چکیده

While most molecular reactions involving synthetic oligonucleotides, notably PCR and DNA sequencing, are carried out in liquid phase, a rapidly growing number of important applications utilize synthetic oligonucleotides attached to solid supports. In general, the purpose for using a solid support-attached oligonucleotide is to capture something, hold on to it, enrich it, and identify and/or purify it. The major class of solid support-attached oligonucleotides, DNA oligonucleotide microarrays, provide a powerful tool for analysis of gene expression. The most generally accessible approach for producing oligonucleotide microarrays is to synthesize the individual oligonucleotides separately prior to immobilization on the solid surface. In this case the oligonucleotide is modified with a functional group that allows attachment to a reactive group on a solid surface. Another important class is the micro-bead-attached oligonucleotide.

Strategies for Attaching Oligonucleotides to Solid Support

Many important molecular applications, such as DNA oligonucleotide arrays, utilize synthetic oligonucleotides attached to solid supports. The most accessible approach for producing an oligonucleotide microarray is to synthesize individual oligonucleotides and subsequently immobilize them to a solid surface. For this immobilization to take place, the oligonucleotides must be modified with a func...

New carbamate supports for the preparation of 3'-amino-modified oligonucleotides.

A novel approach for the preparation of oligonucleotides carrying amino groups at the 3'-end is described. Several CPG supports having aminoalkyl groups and 3'-amino-2',3'-dideoxynucleosides linked through base-labile carbamate linkages such as 2-(2-nitrophenyl)ethoxycarbonyl and fluorenylmethoxycarbonyl were prepared using two different strategies. These supports are compatible to the standard...

Hydrophobic affinity chromatography of nucleic acids and proteins.

5' tritylated oligonucleotides binding hydrophobically to low trityl cellulose/sepharose (< 15 microMTr/ml) retain their hydrogen-bonding specificities for complementary sequences. This, constitutes a novel mode of attaching affinity ligands to solid supports, is more convenient than existing methods, and proceeds with 100% yield. The salt, dielectric constant and temperature dependence of thes...

Porous silicon-based nanostructured microparticles as degradable supports for solid-phase synthesis and release of oligonucleotides

We describe the preparation of several types of porous silicon (pSi) microparticles as supports for the solid-phase synthesis of oligonucleotides. The first of these supports facilitates oligonucleotide release from the nanostructured support during the oligonucleotide deprotection step, while the second type of support is able to withstand the cleavage and deprotection of the oligonucleotides ...
